for Histone H4 tetra-acetyl antibody (pAb)Background
Histone H4 is one of the core components of the nucleosome. The nucleosome is the smallest subunit of chromatin and consists of 147 base pairs of DNA wrapped around an octamer of core histone proteins (two each of Histone H2A, Histone H2B, Histone H3 and Histone H4). Chromatin is subject to a variety of chemical modifications, including post-translational modifications of the histone proteins and the methylation of cytosine residues in the DNA. Reported histone modifications include acetylation, methylation, phosphorylation, ubiquitylation, glycosylation, ADP-ribosylation, carbonylation and SUMOylation; these modifications play a major role in regulating gene expression.
Lysine N-acetylation is a dynamic, reversible and tightly regulated protein and histone modification that plays a major role in chromatin remodeling and in the regulation of gene expression in various cellular functions. Acetylation of histone H4 occurs at several different lysine positions in the histone tail, and is performed by Histone Acetyltransferases (HATs) such as Hat1 or Gcn5. Acetylation of histones is often associated with transcriptional activation.
Fluorescent Secondary Antibodies:
To produce superior results with our primary antibodies, Active Motif has developed a variety of high-quality secondary antibodies. The conjugates have been validated in ELISA-type assays, immunofluorescence and in new high resolution fluorescence microscopy techniques (e.g. STED).

for Histone H4 tetra-acetyl antibody (pAb)Immunogen
This antibody was raised against peptide containing the amino terminal region of Tetrahymena histone variant hv1 acetylated at multiple lysines.
for Histone H4 tetra-acetyl antibody (pAb)Application Notes
This Histone H4 tetra-acetyl antibody (H4Ac) has been validated for use in Western blotting (0.5 - 2 µg/ml) and immunofluorescence (1 - 5 µg/ml). It primarily recognizes tetra-acetylated histone H4, secondarily recognizes tri-acetylated forms of histone H4 and faintly recognizes histone H2A acetylated at multiple lysines. The sequence of the immunogen from Tetrahymena hv1 has a high degree of homology to histone H4 in higher eukaryotes. Because this modification is conserved from yeast to human, a wide range of reactivity is expected.
This antibody has not been validated in ChIP, but ChIP-validated antibodies are available for this target: 39179.
