for MeCP2 antibody (pAb)Background
Methylation of mammalian DNA has long been recognized to play a major role in a number of cellular functions such as development and control of gene expression. It is generally associated with the repressive chromatin state. The complex series of events leading to this repressive state involve the coordinated regulation of DNA methyltransferases and two other groups of proteins called the Methyl-CpG binding proteins (MBD proteins) and the Kaiso family of proteins. The MBD family of proteins include MeCP2, MBD1, MBD2, MBD3 and MBD4. MeCP2 (methyl-CpG binding protein 2) can repress transcription by recruiting the Sin3 complex including histone deacetylases (HDACs). MeCP2 is distributed throughout the nucleus in human cells.
for MeCP2 antibody (pAb)Immunogen
This MeCP2 antibody was raised against a mixture of synthetic peptides corresponding to amino acid residues 11-25 and 181-195 of human MeCP2.
for MeCP2 antibody (pAb)Application Notes
This MeCP2 antibody has been validated for use in Western blotting (1 - 2 µg/ml dilution). For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.
for MeCP2 antibody (pAb)Buffer
PBS containing 0.02% sodium azide. Sodium azide is highly toxic.
for MeCP2 antibody (pAb)Positive Control
Active Motif's HeLa whole-cell extract (Catalog No. 40050) can be used as a positive control for MeCP2 antibody.
MeCP2 antibody (pAb) tested by Western blot.
The analysis was performed using HeLa whole-cell extract and MeCP2 pAb at a 2 µg/ml dilution.
for MeCP2 antibody (pAb)Storage
Antibodies in solution can be stored at -80°C for 2 years. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
