Nuclear lamins are intermediate filament proteins that are the major structural component of the nuclear lamina on the inner surface of the nuclear envelope. Lamins A and Lamins C are splice variants of the Lamin A gene. Lamin A/C (CDCD1, LMN1, EMD2) expression is a hallmark of embryonic stem cell differentiation. In addition to adding structural integrity to the nucleus, lamins contribute to the makeup of the nuclear matrix. Lamins also help organize interphase chromatin through interactions with several chromatin proteins, including histones and Lap2, such that alteration in lamin organization results in disruption of DNA replication, transcription and RNA processing.

This Lamin A/C antibody has been validated for use in Western blotting (0.5 - 2.0 µg/ml dilution), immunofluorescence (5 - 10 µg/ml dilution) and immunoprecipitation (2 - 5 µg per IP). Both lamin A and lamin C are recognized by this sera, with lamin A (the slower migrating band on Western blot) sometimes appearing as a doublet. Addition of 0.05% Tween 20 in blocking buffer and primary antibody incubation buffer may help detection by Western blot.

Bridging Antibody to Improve ChIP, MeDIP and IP

Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use Lamin A/C antibody (mAb).
