Background

NF-1A (NF-1, Nuclear factor-1) is a ubiquitous 47 kDa dimeric DNA-binding protein whose recognition sequence, TGG(C/A)N(5)GCCAA, is found in the genomes of a number of DNA viruses. The same sequence occurs frequently in the human genome. The NF-1 protein stimulates initiation of adenovirus DNA replication in vitro and is capable of stimulating the transcription of genes in cooperation with other factors, such as the estrogen receptor. The Nuclear factor-1 (NF-1) family of transcription factors are encoded by the genes NF-1A, NF-1B2, NF-1C and NF-1X, and feature a highly conserved N-terminal DNA binding region and a variable C-terminal transcriptional activation or repression domain. NF-1 binding controls the expression of many target genes, including insulin, TGF-β, nucleotide, steroid hormone and vitamin synthetic genes, TNFα and FSH. NF-1 transcripts can be differentially spliced and the resultant protein variants homo- and heterodimerize, allowing for multiple potential transcriptional activation and repression activities.
