for Suz12 antibody (mAb) (Clone 2A09)Background

Suz12 (Suppressor-of-Zeste 12 protein, also designated as ChET9, JJAZ1 protein) is a component of the Polycomb group complexes 2, 3 and 4 (PRC2, PRC3 and PRC4), which also contain the histone methyltransferase EZH2, EED (Embryonic Ectoderm Development protein) protein and the histone-binding proteins RbAP46 and RbAP48. PRC2 is responsible for di- and trimethylation of histone 3 at lysine 27 (Histone H3 dimethyl Lys27 and Histone H3 trimethyl Lys27), which leads to transcriptional repression of the target genes. Suz12 is a nuclear protein belonging to the VEFS family. Suz12 contains a zinc-finger binding motif and a VEFS box, which is required for the interaction between Suz12 and EZH2.

This Suz12 antibody was raised against a recombinant protein corresponding to full length human Suz12.

for Suz12 antibody (mAb) (Clone 2A09)Application Notes

This Suz12 antibody has been validated for use in chromatin IP (2.5 - 10 µg per ChIP), immunofluorescence, immunohistochemistry and immunoprecipitation (2 - 5 µg per IP). See references for more information.

This antibody is not recommended for use in Western blotting. Use 39639 instead.

Active Motif's HeLa nuclear extract (Catalog No. 36010) can be used as a positive control for Suz12 antibody.

Chromatin IP Assays

Active Motif has a variety of Chromatin Immunoprecipitation Kits for every need.
Please visit our Chromatin IP Page to learn about ChIP products designed for use with Suz12 antibody (mAb).

Bridging Antibody to Improve ChIP, MeDIP and IP

Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use Suz12 antibody (mAb).

Compatible Secondary Antibodies

Active Motif has developed a variety of high-quality secondary antibodies for most applications, with anti-rabbit and anti-mouse conjugates to a broad line of high-quality fluorescent molecules, as well as horse radish peroxidase (HRP). Visit our Secondary Antibody Conjugates page to find the perfect secondaries for immunofluorescence experiments with Suz12 antibody (mAb).

Suz12 antibody (mAb) tested by ChIP.

Suz12 antibody (mAb) tested by ChIP.
Chromatin IP performed using the ChIP-IT® Express Kit (Catalog No. 53008) and HeLa chromatin (1.5 x 10^6 cell equivalents per ChIP) using 10 µg of Suz12 antibody or the equivalent amount of rabbit IgG as a negative control. Real time, quantitative PCR (RT-qPCR) was performed on DNA purified from each of the ChIP reactions using a primer pair specific for the Myt1 gene promoter. Data are presented as Fold Enrichment of the ChIP antibody signal versus the negative control IgG using the ddCT method.
