TRANCE / RANKL – Tumor Necrosis Factor-Related Activation-induced Cytokine, also known as Receptor Activator of NFκB Ligand is a member of the tumor necrosis factor (TNF) cytokine family and is a ligand for osteoprotegerin (OPG). TRANCE/RANKL functions as a key factor for osteoclast differentiation and activation, is a key survival factor for dendritic cells and is involved in the regulation of T cell-dependent immune response. Overexpression of this protein leads to an increase of osteoclastogenesis and bone loss. This protein may also have a role in regulating apoptosis, by activating the anti-apoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and tumor necrosis factor receptor-associated factor 6 (TRAF6).

This TRANCE/RANKL antibody has been validated for use in Western blotting (4 - 6 µg/ml dilution), and reported to work in immunohistochemistry and flow cytometry. For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Individual optimization may be required.
