Immunogen

This LXR-β antibody was raised against a peptide in the N-terminal region of human LXR-β.

Buffer

Purified rabbit IgG in PBS with 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

 

LXR-β pAb tested by ChIP-Chip.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with chormatin from 2.5 million primary human liver cells. ChIP DNA was amplified by WGA, labeled and hybridized to a human tiling array. The image is zoomed in to show LXR-β binding at a known LXR binding site in the RARB promoter.

LXR-β pAb tested by ChIP-qPCR.
Chromatin immunoprecipitation (ChIP) was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 25 µg of human liver chromatin and 10 µl of LXR-β antibody. ChIP DNA was used in qPCR with the negative control primer pairs and primers against the known LXR binding site in the RARB promoter. Data are presented as Binding Events Detected per 1000 Cells using Active Motif's Epigenetic Services normalization scheme which accounts for primer efficiency and the amount of chromatin used in the ChIP reaction.

Background

LXR-β (Liver X receptor beta, NR1H2, NER, UNR) is an orphan nuclear receptor and a member of the NR1 receptor family. The LXR receptors (including LXR-α and LXR-β ) bind oxysterols and play key roles in maintaining cholesterol homeostasis in macrophages, primarily by regulating multiple components of the reverse cholesterol transport. They also are potent inhibitors of inflammation and are capable of repressing cytokine and chemokine production by Toll-like receptor (TLR)-activated macrophages. LXR-β is more widely expressed than LXR-α, particularly in the brain.

Positive Control

Active Motif's HeLa whole-cell extract (Catalog No. 40050) can be used as a positive control for LXR-β antibody.

qPCR Primers

Active Motif's Human Negative Control Primer Set 1 (Cat. No. 71001) has been validated for performing qPCR and endpoint PCR when this antibody has been used for ChIP on human samples.
