Applications
Application Notes
Applications Validated by Active Motif:
ICC/IF: 1:500 dilution
WB*: 1:500 - 1:2,000 dilution
This antibody is also available as an AbFlex® engineered recombinant antibody. For details on the corresponding AbFlex Recombinant Antibody, see Catalog No. 91231.
*Note: many chromatin-bound proteins are not soluble in a low salt nuclear extract and fractionate to the pellet. Therefore, we recommend a High Salt / Sonication Protocol when preparing nuclear extracts for Western blot.
Immunogen
This Nucleolin antibody was raised against a salt-extracted nuclear matrix preparation from MDCK cells.
Buffer
Concentrated culture supernatant.

Staining of HeLa cells with Nucleolin mAb (Clone 3G4B20) (1:500 dilution, top panel) and DAPI (middle panel), and a merge of both images (bottom panel).
Nucleolin mAb (Clone 3G4B20) tested by Western blot.
HeLa whole-cell extract (15 µg/lane) probed with Nucleolin mAb (Clone 3G4B20) (1:2,000 dilution).
Background
Nucleolin (NCL, C23) is an abundant and ubiquitously expressed protein that is a major component of the nucleolus. Nucleolin seems to be over-expressed in highly proliferative cells and is involved in many aspects of gene expression: chromatin remodeling, DNA recombination and replication, RNA transcription by RNA polymerase I and II (RNA pol II), rRNA processing, mRNA stabilization, cytokinesis and apoptosis. Its role as a histone chaperone has recently been identified. It is involved in the synthesis and maturation of ribosomes, and is located mainly in dense fibrillar regions of the nucleolus.
Positive Control
Active Motif's HeLa whole-cell extract (Catalog No. 40050) can be used as a positive control for Nucleolin antibody (Clone 3G4B20).
Storage
Some products may be shipped at room temperature. This will not affect their stability or performance. Avoid repeated freeze/thaw cycles by aliquoting items into single-use fractions for storage at -20°C for up to 2 years. Keep all reagents on ice when not in storage.
