PARP-2 encodes a chromatin-associated enzyme, a poly-ADP ribosyl transferase, which modifies various nuclear proteins by poly-ADP ribosylation. The modification is dependent on DNA and is involved in the regulation of various important cellular processes such as differentiation, proliferation and tumor transformation. It also plays a role in the regulation of the molecular events involved in the recovery of cells from DNA damage. PARP-2 is cleaved by caspase enzymes during the process of apoptosis.

Addition of 0.05% Tween-20 in blocking buffer and primary antibody incubation buffer is recommended to aid in detection by Western blot.

Lane 1: U2OS nuclear extract (20 µg). Lane 2: A549 whole cell extract(20 µg). Lane 3: HepG2 whole cell extract (20 µg). Lane 4: Jurkat cell nuclear extract (20 µg). These were probed with PARP-2 antibody at a dilution of 1:500.
