for HIRA antibody (mAb) (Clone WC15)Background

HIRA (DGCR1, TUP1) is a histone chaperone that preferentially places the variant Histone H3.3 in nucleosomes, replacing canonical Histone H3 during the cell cycle. HIRA is required for the periodic repression of histone gene transcription during the cell cycle and when complexed with ASF1α is involved in the formation of the senescence-associated heterochromatin foci.

for HIRA antibody (mAb) (Clone WC15)Immunogen

This HIRA antibody was raised against a GST-fusion protein containing residues 421-729 of human HIRA.

for HIRA antibody (mAb) (Clone WC15)Application Notes

This HIRA antibody has been validated for use in immunoprecipitation (5 µg per IP).

for HIRA antibody (mAb) (Clone WC15)Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 32 mM glycine, 0.07 mM EDTA, 30% glycerol, and 0.035% sodium azide.

for HIRA antibody (mAb) (Clone WC15)Positive Control

Active Motif's HeLa acid extract (Paclitaxel treated) (Catalog No. 36201) can be used as a positive control for HIRA (Clone WC15) antibody.

Bridging Antibody to Improve ChIP, MeDIP and IP

Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use HIRA antibody (mAb).

HIRA mAb (Clone WC15) tested by Western blot.
5 µg of HIRA mAb (Clone WC15) was used to immunoprecipitate HIRA from 500 µg of colcemid-treated U2OS nuclear extract (lane 2). 5 µg of control mouse IgG was also used (lane 1). The immunoprecipitated protein was detected by Western blotting using HIRA mAb (Clone WC15) at a dilution of 5 µg/ ml.

for HIRA antibody (mAb) (Clone WC15)Storage

Antibodies in solution can be stored at -20°C for 2 years. Avoid repeated freeze/thaw cycles and keep on ice when not in storage.
