for MMSET / WHSC1 antibody (mAb) (Clone 29D1)Background
MMSET (Multiple Myeloma SET domain-containing protein) / WHSC1 (Wolf-Hirschhorn Syndrome Candidate 1; also known as NSD2, Nuclear Receptor binding SET domain protein 2; TRX5, Trithorax 5) is a SET domain-containing histone methyltransferase specific for lysine 36 of histone H3. The MMSET / WHSC1 gene is deleted in Wolf-Hirschhorn syndrome, a disorder characterized by developmental defects and mental retardation. In 15% of patients with Multiple Myeloma (MM), a chromosomal translocation leads to MMSET / WHSC1 gene being aberrantly placed under the control of the IgH intronic µ enhancer and expressed at very high levels. This event is apparently a key component in cellular transformation leading to MM. Reduction in MMSET / WHSC1 expression in a myeloma cell line suppressed its growth, suggesting a role for MMSET in maintaining the transformed state.

This MMSET / WHSC1 antibody was raised against a recombinant protein corresponding to amino acids 1-647 of human MMSET-I.
for MMSET / WHSC1 antibody (mAb) (Clone 29D1)Application Notes
This MMSET / WHSC1 antibody has been validated for use in chromatin IP (2 - 5 µg per ChIP), immunofluorescence (2 - 5 µg/ml), immunoprecipitation (2 - 5 µg per IP), and Western blotting (0.2 - 1 µg / ml).
Multiple transcripts and splice variants exist for MMSET. This antibody recognizes two major bands: MMSET-I at 88 kDa and MMSET-II at 157 kDa. See references for more information.

Chromatin IP Assays
Active Motif has a variety of Chromatin Immunoprecipitation Kits for every need.
Please visit our Chromatin IP Page to learn about ChIP products designed for use with MMSET / WHSC1 antibody (mAb).
Bridging Antibody to Improve ChIP, MeDIP and IP
Some isotypes of mouse IgG antibodies do not bind well to protein G-conjugated agarose/magnetic beads, which are commonly used in ChIP, MeDIP and IP experiments. Capture efficiency can be improved by including an anti-mouse IgG bridging antibody because it binds with a strong affinity to both the protein G beads and the mouse IgG antibody. By increasing the affinity of the mouse primary antibody for the protein G bead, the Bridging Antibody for Mouse IgG can help improve the results of all ChIP, MeDIP and IP experiments that use MMSET / WHSC1 antibody (mAb).
Compatible Secondary Antibodies
Active Motif has developed a variety of high-quality secondary antibodies for most applications, with anti-rabbit and anti-mouse conjugates to a broad line of high-quality fluorescent molecules, as well as horse radish peroxidase (HRP). Visit our Secondary Antibody Conjugates page to find the perfect secondaries for immunofluorescence experiments with MMSET / WHSC1 antibody (mAb).
