Increasing index base diversity improves sequencing accuracy and cluster identification on Illumina platforms—especially for low-complexity libraries. It serves as a technical constant to monitor sequencing quality and enhance nucleotide diversity and is particularly beneficial for low-complexity libraries.
Diversi-Phi Indexed PhiX is a non-denatured double-stranded DNA PhiX library with all combinations of 8 x 8 bp indexes provided in 10 nMol in 10 mM Tris pH8.0 (10µl). Because Diversi-Phi Indexed PhiX is dual-indexed, it will diversify Read 1, Index 1, Index 2, Read 2.
The index sequences are designed with Hamming distance compatibility so that one or two misread bases will not convert them into another index. The benefits of this color balance in sequencing are:
- Prevents misassignment of samples: barcode errors caused by sequencing mistakes can be detected and corrected, keeping samples correctly separated in the run.
- Error tolerance: the Hamming distance allows for reliable correction of single-base sequencing errors.

Why sequencing diversity matters
Sequencing diversity (or nucleotide diversity) refers to the proportion of AGCT detected in every cycle of a sequencing run. Each base is read as a different color (or color combination, depending on the sequencing chemistry), so if there is too much of one color, detection will not be accurate or even possible. Including Diversi-Phi Indexed PhiX in libraries will increase the diversity to avoid sequencing detection errors.
