Immunogen

This Histone H3 acetyl Lys27 antibody was raised against a synthetic peptide including acetyl-lysine 27 of human histone H3.

Buffer

PBS pH 7.5 containing 30% glycerol. 0.3M NaCl, and 0.035% sodium azide. Sodium azide is highly toxic.

 

Histone H3K27ac antibody (mAb) tested by ChIP-Seq.
ChIP was performed using the ChIP-IT® High Sensitivity Kit (Cat. No. 53040) with 15 ug of chromatin from a human medulloblastoma cell line and 4 µg of antibody. ChIP DNA was sequenced on the Illumina HiSeq and 17 million sequence tags were mapped to identify Histone H3K27ac binding sites. The image shows binding across a region of chromosome 7. Histone H3 acetyl Lys27 mAb tested by dot blot analysis.
Dot blot analysis was used to confirm the specificity of Histone H3 acetyl Lys27 mAb for acetyl-Lys27 histone H3. Acetylated peptides corresponding to the immunogen and related peptides were spotted onto PVDF and probed with the antibody at 0.2 µg/ml. The amount of peptide (picomoles) spotted is indicated next to each row.

Histone H3 acetyl Lys27 mAb tested by Sysmex Corporation using their Super Resolution Microscope HM-1000
Cos7 cells were stained with Histone H3 acetyl Lys27 mAb, with Alexa Fluor 488 (Top/Green) conjugated to anti-mouse IgG as a secondary antibody. HeLa cells were also stained with Histone H3 acetyl Lys27 mAb, with Chromeo 488 (Bottom/Green) conjugated to anti-mouse IgG (ActiveMotif) as a secondary antibody. Images were obtained at an exposure time of 30 milliseconds.
