Immunogen

This HP1 gamma phospho Ser93 antibody was raised against a peptide containing phospho Ser93 of human HP1 gamma.

Buffer

Purified IgG in 70 mM Tris (pH 8), 105 mM NaCl, 31 mM glycine, 0.07 mM EDTA, 30% glycerol and 0.035% sodium azide. Sodium azide is highly toxic.

 

HP1 gamma phospho Ser93 pAb tested by Western blot.
HeLa nuclear extract (20 µg per lane) was probed with HP1 gamma phospho Ser93 pAb (1:500 dilution). Lane 1: no treatment. Lane 2: cells treated with colcemid.

HP1 gamma phospho Ser93 pAb tested by dot blot analysis.
Dot blot analysis was used to confirm the specificity of HP1 gamma phospho Ser93 pAb for HP1 gamma phospho Ser93. Phosphorylated peptides corresponding to the immunogen and related peptides were spotted onto PVDF and probed with HP1 gamma phospho Ser93 pAb at 1:5,000. The amount of peptide (picomoles) spotted is indicated next to each row.

Lane 1: unmodified HP1 gamma peptide.
Lane 2: phospho Ser93 HP1 gamma peptide.
Lane 3: phospho Ser92 HP1 alpha peptide.
Lane 4: phospho Ser89 HP1 beta peptide.

Background

Heterochromatin Protein 1 gamma (HP1 gamma) is one of the mammalian homologues of Drosophila HP1, first identified through its role in the maintenance of regions of the genome in a heterochromatic state. HP1 gamma contains a Chromo domain and a Chromo shadow domain. Through the Chromo domain, HP1 proteins are recruited to regions of chromatin that exhibit methylation of lysine 9 of histone H3. Phosphorylation of serine 93 of HP1 gamma increases during mitosis.
