Background

YTHDC2 (YTH Domain Containing 2), is a member of the DEAH (Asp-Glu-Ala-His) subfamily of proteins, part of the DEAD (Asp-Glu-Ala-Asp) box family of RNA helicases. It specifically recognizes and binds to N6-methyladenosine (m6A), a common modified RNA nucleotide that is enriched in the stop codons and 3’ UTRs of eukaryotic mRNAs and some non-coding RNAs and plays a role in the efficiency of mRNA splicing, processing and stability. YTHDC2 may be associated with susceptibility to pancreatic cancer in human patients, and knockdown of this gene resulted in reduced proliferation in a human liver cancer cell line.

Recombinant YTHDC2 protein was expressed in a baculovirus expression system as the full length protein (accession number NP_073739.3) with an N-terminal Flag tag. The molecular weight of the protein is 161.5 kDa.

Recombinant YTHDC2 binding assay
3 µM biotinylated ssDNA m6A oligos (GTTGG/m6A/CTT) were incubated with different concentrations of YTHDC2 protein in a 10 µl system containing 50 mM HEPES-NaOH pH 7.4, and 0.1% BSA for 1 hour, then 10 µl FLAG antibody and SA-XL665 mixture (each 1:100 dilution reaction buffer) was added to each reaction system and incubated for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.

Storage
