Background

HeLa acid extract (PTX treated) was prepared under acidic conditions from cells harvested from a cell culture of the HeLa human epithelial carcinoma cell line that was treated with PTX. Acid extraction is performed under conditions that are optimized for solubility of histone proteins.

PTX is one of a class of diterpenoids known as taxoids that have been shown to be effective chemotherapeutic agents in the treatment of solid tumors in humans. The antineoplastic effect is brought on largely by the ability of taxoids to promote stabilization of tubulin polymerization and thus protect microtubules from disassembly. PTX treatment of HeLa cells is cytotoxic and induces mitotic arrest and apoptotic cell death.

The HeLa cell line was originally derived from cells obtained in 1951 from Henrietta Lacks, a cervical cancer patient who eventually died from her cancer. The HeLa cell line is one of the oldest and most commonly utilized immortal cell lines in scientific research. These cells are highly prolific and easily sustainable, allowing for flexibility in their application in various fields of scientific research, including cancer biology, virology, and human disease. To date, over 60,000 publications across various disciplines have referenced the use of HeLa cells.

Application Notes

Acid extracts are commonly utilized for isolation and purification of histones. HeLa acid extract (PTX treated) is specifically recommended for analysis of the mechanisms governing PTX cytotoxicity, particularly in relation to 1) microtubule assembly and motility, 2) mitotic inhibition, 3) cell survival, and 4) cancer therapies.

Extract Composition

HeLa acid extracts are supplied in dilution buffer (PBS supplemented with 0.5 mM DTT and 2 mM PMSF). Cells were cultured in medium containing 0.1 µg/ml PTX for 20 hours immediately prior to harvesting.

Quality Control

Acid extracts have been quality control tested by Western blot.

Storage

To ensure stability, extracts should be stored at -80°C.

We recommend aliquoting the extracts into single-use fractions and then storing them at -80°C. This eliminates repeated freeze/thaw cycles.
